The dead man was assaulted in an alleyway near to the Royal Court Hotel in Lowestoft at 10.25pm on Wednesday.
The victim was taken to the James Paget Hospital in Gorleston with serious head injuries, but police said last night he had died.
Two males - including a 16-year-old boy - were being quizzed by detectives at separate Suffolk police stations on suspicion of murder today.
A third man - a 23-year-old from Lowestoft - has now been arrested on suspicion of murder and taken into custody at Halesworth police station.
It is believed the dead man lived in the Lowestoft area.
