If they can overcome the London Two South West side, Mark Bolton's men would be just two matches away from Twickenham.
Tottonians are currently second in their league and, like Sudbury, are enjoying a successful season.
The winners of the tie will play against either Saffron Walden or Tonbridge Juddian before going into a semi-final draw with the winners of the Midlands, Northern and South West Intermediate Cups.
