The monarch has a trip planned to the US state at the beginning of next month as part of a royal visit to America.
A Buckingham Palace spokeswoman said: “The Queen was shocked and saddened to hear of the news of the shooting in Virginia.
“She will be kept informed.”
The Queen and the Duke of Edinburgh are to pay a two-day visit to Virginia on May 3 and 4 where the monarch will commemorate the 400th anniversary of the Jamestown Settlement.
It will be her first state visit to America for 16 years.
